Polyhydroxy Butyrate Using Biogas Slurry

Abstract
As the world continues to address the plastic waste challenges, PHB emerges as a game-changing solution. Its adoption and further development can significantly contribute to reducing plastic waste, mitigating environmental harm and promoting a more sustainable future. This research aims at the production of polyhydroxybutyrate using biogas slurry. Streak plate method was used for obtaining the purified strain. The primary and secondary screening of PHB producing bacteria was carried out using biochemical tests such as catalase test, indole test, motility test etc. The research results shown that PHB 1, PHB 3, PHB 5, PHB 6, PHB 7, PHB 8, PHB 9, PHB 10 shows strong staining with Sudan B black while PHB 2, PHB 4 exhibits weak staining. All isolates were Gram-positive, all isolates were rod-shaped, , all isolates except PHB9 were oxidase positive, all isolates except PHB5 produced hydrogen sulphide, all isolates utilized glucose, sucrose utilization varied, with PHB2 and PHB6 negative and the others showing mixed results. Only PHB5 utilized lactose, citrate utilization was variable, with PHB2, PHB6, and PHB8 positive, Indole None of the isolates produced indole, all isolates were capable of nitrate reduction, Voges Proskauer; All Bacillus species tested positive, indicating acetoin production. Xylose, Inositol, Arabinose, Galactose were all tested positive showing the ability to utilize these sugars. All species tested negative Methyl Red, suggesting they do not produce stable acids from glucose fermentation. All species were catalase positive, indicating the presence of the catalase enzyme. Our finding shows that PHB is a groundbreaking biopolymer that holds immense potential for transforming plastic industry.
