The Protective Effects of Datura Stramonium Against Cadmium-Induced Oxidative Stress in the Testis of Adult Male Wistar Rats

Abstract
Oxidative stress is a major factor in cadmium-induced testicular toxicity, leading to disrupted antioxidant systems and cellular damage. Previous studies have shown that cadmium significantly alters oxidative stress markers such as glutathione (GSH), oxidized glutathione (GSSH), glutathione peroxidase (GPx), and glutathione S-transferase (GST), which are critical in detoxifying reactive oxygen species (ROS) and maintaining cellular balance. Datura stramonium, a medicinal plant with known antioxidant properties, has been highlighted in the literature for its potential to combat oxidative damage, but its specific effects on testicular health in the context of cadmium exposure remain underexplored. This study investigates the biochemical effects of Datura stramonium on cadmium-induced oxidative stress in the testes of adult male Wistar rats. Thirty rats were divided into six groups: a control group, a cadmium-only group, and groups treated with Datura stramonium extract, cadmium combined with Datura stramonium, and cadmium with Vitamin E. Cadmium was administered intraperitoneally at 30 mg/kg body weight, while Datura stramonium extract was given at 100 mg/kg and 200 mg/kg body weight for 21 days. Blood and testicular tissue samples were analyzed to assess changes in GSH, GSSG, GPx, and GST levels. The study confirms that cadmium exposure induces oxidative stress by reducing GSH, GPx, and GST levels while increasing GSSH. The administration of Datura stramonium improved antioxidant enzyme activity and glutathione balance, indicating its potential as a natural protective agent against cadmium-induced testicular toxicity. While these findings are promising, further studies are needed to establish its safety, optimal dosage, and long-term effects on reproductive health.
