Investigations of the Effect of Dapagliflozin on Motor Coordination and Neuroxidative Stress in Haloperidol Induced Parkinsonism in Wistar Mice

Abstract
Dapagliflozin is a member of Sodium-glucose co-transporter 2 (SGLT 2) inhibitors which are relatively new antidiabetic drug with antioxidant and anti-inflammatory properties. Therefore, this study aimed to investigate whether Dapagliflozin have a neuroprotective effect in parkinsonism model.
Thirty-six Wistar mice were randomized into groups. The first one (control group) received distilled water (10mL/kg P. O). The second group received haloperidol (HAL) (0.1 mg/kg/, I. P) only, whereas the third, fourth, fifth and sixth groups received trihexyphenidyl ((THP) (2 mg/kg, P. O), Dapagliflozin (DAPA) (2.5 mg, 5 mg and 10 mg/kg/day, orally), respectively followed by HAL (0.1 mg/kg/, I. P) daily. All drug administrations were continued daily for 21 days.
Rotarod test was done on day 1 and day 21 of administration. On day 21, mice were sacrificed, brain tissues were collected for analysis.
HAL significantly caused motor impairment and elevated oxidative stress markers. Groups of (DAPA + HAL) significantly improved motor coordination and alleviated oxidative stress parameters.
In both low and high doses, DAPA produced a neuroprotective effect against the HAL mice model, with the high dose inducing a more significant effect.
