The Impact of Curcumin Supplement on Renal Tissue Inflammatory Markers in Alloxan-Induced Male Diabetic Wistar Rats

Abstract
Background: Diabetes mellitus is a chronic metabolic disorder which is characterized by a high blood sugar level known as hyperglycemia, which often leads to various complication, including renal dysfunction which Inflammation plays a pivotal role in. Interleukin 6(IL 6) and tumor necrosis factor alpha (TNF-α) contribute to renal injury through the promotion of oxidative stress, fibrosis and apoptosis. This study aims to evaluate the effect of curcumin supplement on the levels of IL 6 and TNF- α as renal inflammatory markers in alloxan-induced diabetes rats.
Methodology: a total number of forty-five (45) matured Wistar of average weight 160 to 200g were used for this study. They were divided into five group (A to E) nine animal in each group. Group A the control received normal feed and water. Group B (Alloxan only), Group C (Alloxan and metformin) and Group D (Alloxan and Curcumin) were administered 150mg\kg of alloxan intraperitoneally to induce diabetes. Their blood glucose level was checked after 72hours to confirm diabetes. After confirming the animals diabetics, animals in group C were treated with metformin (100mg\kg) daily for 4 weeks. After 4weeks of treatment, the animal were fasted overnight for 10 to 12 hours. The experimented animals were sacrificed and kidney were harvested and homogenized. The kidney homogenates were used for the determination of renal inflammatory markers. The data obtained were analyzed using graph pad prism 5(Graph Pad Software Inc., San Diego, CA).
