Effect of Chronic Sleep Deprivation on Markers of Inflammation in Rats With Acetic Acid Induced Peptic Ulcer

Abstract
Sleep plays a vital role in maintaining physiological homeostasis, and its deprivation has been linked to several pathological conditions. This study investigated the effects of chronic sleep deprivation on inflammatory markers in male Wistar rats with acetic acid-induced peptic ulcers. A total of 110 rats were divided into ten experimental groups, including control, ulcer control, sleep-deprived, and combined ulcer plus sleep-deprived groups. Sleep deprivation was induced using the gentle handling technique, while peptic ulcers were created using 20% acetic acid. Various behavioral, biochemical, and histological analyses were conducted to evaluate inflammation and tissue damage. The findings revealed that chronic sleep deprivation significantly elevated inflammatory biomarkers such as Tumor Necrosis Factor-alpha (TNF-α) and Nuclear Factor kappa B (NF-κB) in both ulcerated and non-ulcerated rats. Histological analysis showed increased mucosal erosion and inflammatory infiltration in sleep-deprived ulcer groups compared to controls. These results indicate that chronic sleep deprivation exacerbates inflammatory responses and worsens gastric mucosal injury in peptic ulcer conditions. Conclusively, the study highlights a strong interaction between sleep deprivation and gastrointestinal health, suggesting that adequate sleep is crucial in preventing or mitigating inflammation-associated gastrointestinal disorders.
