Isolation and Characterisation of Lipase Producing Strains Isolated from Termite Moth and Oily Soil from a Mechanic Workshop

Abstract
Alkaline lipase production is of significant industrial interest due to its applications in detergents, bioremediation, and the synthesis of biodiesel. However, the optimization of production processes remains a challenge, primarily due to the limited understanding of the factors influencing enzyme yield and activity. This study aimed to isolate and characterize the bacteria strain that is responsible for the production of Lipase. The soil sample were collected from termite moth and oily sands around mechanic workshop into the laboratory, ten fold serial dilution was carried out to reduce the microbial load, and then transferred to nutrient medium in a Petri dish and it was incubated. After then the isolate observed with clearance zone of 10mm was selected. Colonial morphological characteristics was employed, the selected bacterial isolate was characterized using the following colonial characters such as colony, shape, size, color, optic, surface texture, margin, and elevation. Microscopic morphological characteristics which shows whether the isolate is gram positive or negative. Biochemical test used to identify bacterial species by differentiating them on the basis of biochemical activities. The wet biomass and cell growth of the lipase isolates were determined using oven drying and spectrophotometric methods. The results shows that termite moth has the highest count of 27.9 and 13.2 while oily soil has its own count of 6.2 and 4.8 respectively. The result shows that the microorganism isolated was Bacillus spp. The result indicates that at 72 hours the best biomass was obtained. The result indicates that at 144 the highest optical value of 3.000 was obtained. These findings have important implications for industrial applications, as they provide a more efficient and cost effective method for producing lipase.
