The formulation of cost-effective media using locally sourced materials for the mass production of bacterial endospores, specifically Bacillus cereus, presents an innovative approach to microbial cultivation. This study aimed to develop an affordable and efficient culture medium by utilizing agricultural products such as millet, beans, groundnut, sorghum, and sesame. Soil samples were collected from an agricultural field, and Bacillus cereus was isolated through heat treatment, serial dilution, and selective culturing. Biochemical tests, including catalase, citrate, motility, and spore staining, were performed to confirm bacterial identification. Various combinations of carbohydrate-to-protein ratios were tested to determine the optimal growth conditions. The results showed that millet and beans, as well as millet and groundnut, provided the most suitable growth medium, with an observed growth rate of up to 80% at lower dilution levels. This study highlights the potential of locally sourced materials in reducing the cost of bacterial culture media while maintaining efficiency. The findings can be applied in industrial microbiology and agricultural biotechnology for large-scale bacterial endospore production.
