Isolation and Identification of Bacteria Responsible for the Spoilage of Leafy Vegetable (pumpkin Leaf and Bitter Leaf)

Abstract
ABSTRACT
The spoilage of vegetables, particularly bitter leaf (Vernonia amygdalina) and pumpkin leaf
(Telfairia occidentalis), poses significant challenges to food quality and safety. This study
ailned to isolate and characterize the bacterial species responsible for the spoilage of these
vegetables. Samples of spoiled bitter leaf and pumpkin leaf were collected and subjected to
standard microbiological techniques for bacterial isolation. The isolates were then
characterized through a series of cultural, morphological, and biochemical tests, including
Gram staining, catalase, indole, oxidase, and motility tests. Results revealed a diverse range
of bacterial species associated with spoilage. The isolates included Klebsiella spp., Bacillus
aureus, Pseudonwnas spp., and Staphylococcus spp. The study underscores the importance of
implementing effective hygienic practices, temperature control, and antimicrobial treatments
to mitigate bacterial spoilage of vegetables. Further research into the microbial communities
and their spoilage mechanisms is recommended to develop targeted strategies for extending
bitter leaf and pumpkin leaf.
