Effects of Justicia Carnae on Heme Oxygenase Gene Expression, Malondialdehyde, Interleukin6, and Heme Levels in Sodium Meta Bisulfite Induced Wistar Rat

Abstract
ABSTRACT
Background: Sickling and oxidative stress are associated with disruptions in heme metabolism, inflammatory cytokines, and lipid peroxidation, contributing to tissue damage and disease progression. Justicia carnea, widely used in traditional medicine, has gained attention for its potential as a blood-booster in anemic conditions and sickle cell anemia.
Aims and Objectives: This study aimed to investigate the effects of Justicia carnea leaf extract on heme oxygenase gene expression, malondialdehyde (MDA) levels, interleukin-6 (IL-6), and heme levels in sodium metabisulfite-induced Wistar rats.
Method: Twenty-four male Wistar rats were assigned to six groups: (Group 1) normal feed (control), ( group 2) normal feed + sodium metabisulfite, ( group 3) normal feed + sodium metabisulfite + Ciklavit, and ( group 4) normal feed + sodium metabisulfite + folic acid, ( group 5) normal feed + sodium metabisulfite + low dose Justicia carnea, ( group 6) normal feed + sodium metabisulfite + high dose Justicia carnea. All rats were fed with the normal animal feed throughout the period of the experiment. Sodium metabisulfite was administered after two weeks, and treatments with Justicia carnea, Ciklavit, and folic acid were provided for an additional two weeks Expression levels of Heme-Oxygenase, malondiahyde , interleukin-6 and Heme level were quantified..
Results: Results demonstrated that Justicia carnea significantly modulated heme oxygenase gene expression, reduced MDA levels, and downregulated IL-6 expression. Furthermore, the extract restored altered heme levels, showcasing its antioxidative and anti-inflammatory properties.
Conclusion: These findings highlight the therapeutic potential of Justicia carnea leaf extract in mitigating oxidative stress, inflammation, and heme dysregulation, offering a promising natural approach for managing related disorders. Further research is recommended to explore its broader clinical applications.
