Amplification of Alkbi Gene in Diesel Degrading Bacteria from Dumpsite.

Abstract
ABSTRACT
This current study investigated the screening of bacteria isolates from dumpsite soil for degradative
genes.Dump site soil samples were collected under sterilized conditions from Ekiti State
University,Ado-Ekiti,Ekiti State,Nigeria.Morphological characterization of all the pure cultures
on Nutrient Agar and MacConkey Agar showed opaque attributes when observed.The pure culture
of bacteria on Nutrient Agar in sample Dla showed a total number of seventy five colonies;and
some colonies showed circular shape,creamy white colour and flat elevation,but the remaining
colonies showed medium size while the remaining colonies showed small and tiny size.The pure
culture of bacteria on MacConkey Agar in sample D2a showed a total number of fifty three
colonies;some of the colonies showed circular shape,small size,white colour and flat elevation
while the remaining colonies showed circular shape,tiny size,pink colour and flat elevation.Also,
,he bacteria isolates obtained from sample ID:D1Ta,D1S,DIT and D3Ma were found to degrade
hydrocarbons heavily.Meanwhile,the bacteria isolates obtained from sample ID:D3Ma,D2Ma,
D1Sa,D4M and D3Ma were found to degrade hydrocarbons moderately.However,the bacteria
isolates obtained from sample ID:D3Tb,D2Tb and D3Sb were found to degrade hydrocarbons
lightly.At 2%concentration,bacteria strains D8 showed the hydrocarbon reductance level at
0.300nm,followed by bacteria strains D1,D2,D3 and D5 at 0.200nm.At 3%and 5%
concentration,all the bacteria isolates obtained from dumpsites showed hydrocarbon reductance
level at 0.100nm.Meanwhile,at 10%concentration,bacteria strains D7 showed hydrocarbon
reductance level at 0.200nm while others showed hydrocarbon reductance level at 0.100nm.
Sample D5,sample D7 and sample D8 were identified to be Pseudomonas aeruginosa strain
NSL416S ribosomal RNA gene,partial sequence,with accession number OQ975911;Proteus
penneri strain Rizhao_556_116S ribosomal RNA gene,partial sequence,with accession number
MN251918 and Alcaligenes faecalis strain C516S ribosomal RNA gene,partial sequence with
accession number OP316827.It was found that potential bacteria that could degrade diesel would
readily be isolated from hydrocarbon-contaminated soil samples or other natural environments that
have no direct contamination with hydrocarbon residuals.This could be a groundwork indication
for a possible search of potential bacterial isolates for the bioremediation of hydrocarbon-
contaminated environments.
